Feeding and Temperature Responses to Intravenous Leptin Infusion Are Differential Predictors of Obesity Gain in Rats

چکیده
Obesity is frequently associated with leptin resistance. The present study investigated whether leptin resistance in rats is present before obesity develops, and thus could underlie obesity induced by 16 weeks exposure to a liquid, palatable, high-energy diet (HED). Before HED exposure, male Wistar rats (weighing between 330 and 360 g) received i.v. infusions of 20 μg of leptin 2h before dark (appr. 57 μg/kg rat). Relative to saline infusion, this caused a highly variable effect on food intake (ranging between –94 and +129%) with food intake suppression that appeared negatively correlated with HED-induced increases in body weight gain, caloric intake, adiposity, and plasma leptin levels. In contrast, leptin’s thermogenic response was positively correlated to body weight gain linked to weights of viscera, but not to adiposity. Before HED exposure, leptin unexpectedly increased food intake in some rats (fi+, n=8), whereas others displayed the normal reduction in food intake (fi-, n=7). HED exposed fi+ rats had higher plasma leptin levels, retroperitoneal fat pad weight, HED intake, and body weight gain than fiand CHOW rats. These parameters were also higher in HED exposed firats relative to CHOW rats, except for plasma leptin concentrations. It is concluded that leptin’s reduced efficacy to suppress food intake could predict obesity on a HED. An unexpected orexigenic effect of leptin might potentially contribute to this as well.

متن کاملComparison of the Effects of Cuminum Cyminuml and Sibutramine on Weight, Serum Leptin, Glucose and Lipids in Rat
Objectives: Diabetes is one of the most common metabolic diseases in the world. It affects 6.6% of world population and about 3 million individuals in Iran. Nowadays chemical and herbal medicines are widely prescribed to cure obesity. In Iran, cumin (Cuminum Cyminum L.) is a plant used in traditional medicine to cure obesity, and some of the new studies have suggested that cumin has a role in d...
متن کاملLeptin responses to glucose infusions in obesity-prone rats.
The secretion of leptin is dually regulated. In fasting animals, plasma leptin concentrations reflect body fat stores, whereas the incremental leptin response to fasting or refeeding most likely reflects insulin-mediated energy flux and metabolism within adipocytes. Impaired secretion of leptin in either pathway could result in obesity. We therefore measured plasma leptin concentrations in fast...
متن کاملAnti-Obesity, Fat Lowering and Liver Steatosis Protective Effects of Ferula asafoetida Gum in Type 2 Diabetic Rats: Possible Involvement of Leptin
Objective: Control of weight gain is an important strategy in reducing the diabetes incidence. Recently, herbal drugs have been used as a complementary and alternative medicinal care. This study was conducted to determine the effect of Ferula asafoetida on weight gain, fat accumulation, liver steatosis and leptin level. Materials and Methods: All rats of control and treatment groups received d...
متن کاملThe Effect of Peripheral Injection of Leptin on Biomarkers of Aging in Calorie Restricted Rats
Background and Objectives: Calorie restriction (CR) is one of the proven methods of extending lifespan and slowing aging. Leptin is a nutritionally regulated adipokine that has been proposed as a possible key signal in the adaptive responses relevant to CR. Under CR, plasma leptin levels decrease, thus it was hypothesized that leptin administration may counteract CR-induced weight loss. Therefo...
